It probably will - mostly. Unfortunately, this is one of those things where probably and mostly are not nearly good enough, at least for many of us.
I've had a couple of flats with this Prius. One was a finishing nail, that fit your "typical" scenario. The other was a more substantial bolt, that had the tire dead flat in a a mile or two.
Let's be honest here folks. If you don't care whether you have a spare (or repair kit ), your weren't going to use them when needed anyway...and for the rest, doesn't everybody have chocks?
I had large bolt in a tire once. It actually plugged the hole for several miles, and I might have gotten home with it in place if I stayed to slower roads. I've only used the goo in trying to seal a mystery leak, so I can't say how well it works for larger holes. Will work on holes that a plug won't? If not, will it work with in conjunction to a plug? Locally, there was a pothole that was stranding people by taking out both tires on the side. If damage worse than 'typical' is more common were you drive, shouldn't you be carrying two spares, just in case? This all about risk assessment and comfort. In the 30 or so years of personal memory, I have been involved in a half dozen or so flats that have been, or could have been, fixed by a plug kit. One required a side patch. One was that large bolt, which may or may not have fixed with a plug, a professional plug did fix it. And back when I was around 10, we had a tire blow out. Tire durability has improved with time. All new cars have TPMS to alert the driver to slow leaks, and the damage can be addressed before they lead to a blow out. Then there is cell coverage where I drive. So I wouldn't be concerned if the new car I want doesn't have a spare. Other peoples' experiences and local conditions can change that assessment and comfort.
I've had two occasions in my younger life to use a spare, a bent rim and the other was just plain doing stupid stuff. I've had fast leaks that leave your tire flat in hours. I love those because everyone stops by your desk to tell you. At this point in my life I've learned to avoid the big ones and you can't do what they did on the Duke's oh hazard. You can't always safely use a car jack depending on shoulder conditions and traffic. If you plan on doing so carry some wood to stabilize the jack and chock the wheels.

Has the rear wiper but not the 3 door SK. Odd because the brochure says Eco has the 3 door SK but no rear wiper.
Test drove it in all modes, Eco, Power, Normal and EV. Here's a pic of the window sticker and the rear window wiper. Maybe we got a special one.
Quite possibly not Eco. I emailed for a quote on a Two and when we were test driving it, the salesman was showing me the modes. Maybe the Eco is a charge to add the 3 door SK, the wheel covers and to remove the spare and the wiper. They all have Eco mode if this is the case.
